Description

Description: This assay employs a two-site sandwich ELISA to quantitate DNM3 in samples. An antibody specific for DNM3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any DNM3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for DNM3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of DNM3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Dynamin-3 is a member of the dynamin family which possess mechanochemical properties involved in actin-membrane processes, predominantly in membrane budding. DNM3 is upregulated in Sézary's syndrome. By sequencing clones obtained from a size-fractionated brain cDNA library, Nagase et al. (1998) cloned DNM3, which they designated KIAA0820. The deduced protein contains 702 amino acids. RT-PCR ELISA detected very high expression in adult brain and spinal cord and in all specific brain regions examined. Intermediate expression was detected in adult heart, lung, kidney, testis, and ovary and in fetal liver and brain. Low expression was detected in adult liver and pancreas, and little to none was detected in skeletal muscle and spleen
